    Theatre in Towns offers a contemporary perspective on the role of theatre in the cultural life of towns in England. Exploring volunteer-led, professional and community theatres, this book investigates the rich and diverse ways that theatres in towns serve their locality, negotiate their civic role, participate in networks of mutual aid and exchange, and connect audiences beyond their geographical borders. With a geographical focus on post-industrial, seaside, commuter and market towns in England, the book opens questions about how theatre shapes the narratives of town life, and how localism, networks and partnerships across and between towns contribute to living sustainably.     Quantifying air-sea gas exchange using noble gases in a coastal upwelling zone

We have evaluated the performance of four published air-sea gas exchange parameterizations using a five-day time-series of dissolved He, Ne, Ar, Kr, and Xe concentration in Monterey Bay, CA. We constructed a vertical model including surface air-sea gas exchange and vertical diffusion. Diffusivity was measured throughout the cruise from profiles of turbulent microstructure. We corrected the mixed layer gas concentrations for an upwelling event that occurred partway through the cruise. All tested parameterizations gave similar results for Ar, Kr, and Xe; their air-sea fluxes were dominated by diffusive gas exchange during our study. For He and Ne, which are less soluble, and therefore more sensitive to differences in the treatment of bubble-mediated exchange, the parameterizations gave widely different results with respect to the net gas exchange flux and the bubble flux.     Changes in gross oxygen production, net oxygen production, and air-water gas exchange during seasonal ice melt in Whycocomagh Bay, a Canadian estuary in the Bras d\u27Or Lake system

    Sea ice is an important control on gas exchange and primary production in polar regions. We measured net oxygen production (NOP) and gross oxygen production (GOP) using near-continuous measurements of the O2∕Ar gas ratio and discrete measurements of the triple isotopic composition of O2, during the transition from ice-covered to ice-free conditions, in Whycocomagh Bay, an estuary in the Bras d\u27Or Lake system in Nova Scotia, Canada. The volumetric gross oxygen production was 5.4+2.8-1.6 role= presentation \u3e5.4+2.8−1.6 mmol O2 m−3 d−1, similar at the beginning and end of the time series, and likely peaked at the end of the ice melt period. Net oxygen production displayed more temporal variability and the system was on average net autotrophic during ice melt and net heterotrophic following the ice melt. We performed the first field-based dual tracer release experiment in ice-covered water to quantify air–water gas exchange. The gas transfer velocity at \u3e90 % ice cover was 6 % of the rate for nearly ice-free conditions. Published studies have shown a wide range of results for gas transfer velocity in the presence of ice, and this study indicates that gas transfer through ice is much slower than the rate of gas transfer through open water. The results also indicate that both primary producers and heterotrophs are active in Whycocomagh Bay during spring while it is covered in ice

    Relational counselling as a psychosocial intervention for dementia : qualitative evidence from people living with dementia and family members

    Counselling and other psychotherapeutic interventions can be offered to people with dementia and their caregivers, to treat specific conditions or symptoms (e.g. affective disorders such as depression). Psychotherapeutic interventions also offer the opportunity for individuals with dementia and their families/caregivers to engage in psychological support for a wide range of presentations. However, little is known about how those within this demographic who receive these interventions perceive the experience. This study aimed to understand the experiences of individuals with dementia or caring for someone with dementia, before and after a 12-week relational counselling intervention delivered through a third sector organisation within England. Semi-structured interviews were completed with participants (29 pre-intervention and 25 post-intervention). Framework analysis was conducted, with four main themes identified; expectations and outcomes of counselling, emotional impact of life with dementia, appraisals of identity and importance of therapeutic relationship. Participants reported that counselling interventions addressed a range of needs and concerns that they had, enabling them to reassess and reconsider these. Specific training is needed before therapists deliver therapeutic interventions with people with dementia, to ensure that appropriate support is provided for those with more severe cognitive impairment or who may have fluctuating capacity. Future research should explore the experiences of people with dementia and their caregivers, across different counselling modalities, to establish the appropriateness and effectiveness of relational counselling

    Impact of recently upwelled water on productivity investigated using in situ and incubation-based methods in Monterey Bay

The definitive version was published in Journal of Geophysical Research: Oceans 122 (2017): 1901–1926, doi:10.1002/2016JC012306.Photosynthetic conversion of inline image to organic carbon and the transport of this carbon from the surface to the deep ocean is an important regulator of atmospheric inline image. To understand the controls on carbon fluxes in a productive region impacted by upwelling, we measured biological productivity via multiple methods during a cruise in Monterey Bay, California. We quantified net community production and gross primary production from measurements of inline image/Ar and inline image triple isotopes ( inline image), respectively. We simultaneously conducted incubations measuring the uptake of 14C, inline image, and inline image, and nitrification, and deployed sediment traps. At the start of the cruise (Phase 1) the carbon cycle was at steady state and the estimated net community production was 35(10) and 35(8) mmol C m−2 d−1 from inline image/Ar and 15N incubations, respectively, a remarkably good agreement. During Phase 1, net primary production was 96(27) mmol C m−2 d−1 from C uptake, and gross primary production was 209(17) mmol C m−2 d−1 from inline image. Later in the cruise (Phase 2), recently upwelled water with higher nutrient concentrations entered the study area, causing 14C and inline image uptake to increase substantially. Continuous inline image/Ar measurements revealed submesoscale variability in water mass structure and likely productivity in Phase 2 that was not evident from the incubations. These data demonstrate that inline image/Ar and inline image incubation-based NCP estimates can give equivalent results in an N-limited, coastal system, when the nonsteady state inline image fluxes are negligible or can be quantified.Funding for this work was provided by NSF awards OCE-1060840 to R.H.R.     Glucocorticoids regulate mitochondrial fatty acid oxidation in fetal cardiomyocytes

    Abstract: The late gestational rise in glucocorticoids contributes to the structural and functional maturation of the perinatal heart. Here, we hypothesized that glucocorticoid action contributes to the metabolic switch in perinatal cardiomyocytes from carbohydrate to fatty acid oxidation. In primary mouse fetal cardiomyocytes, dexamethasone treatment induced expression of genes involved in fatty acid oxidation and increased mitochondrial oxidation of palmitate, dependent upon a glucocorticoid receptor (GR). Dexamethasone did not, however, induce mitophagy or alter the morphology of the mitochondrial network. In vivo, in neonatal mice, dexamethasone treatment induced cardiac expression of fatty acid oxidation genes. However, dexamethasone treatment of pregnant C57Bl/6 mice at embryonic day (E)13.5 or E16.5 failed to induce fatty acid oxidation genes in fetal hearts assessed 24 h later. Instead, at E17.5, fatty acid oxidation genes were downregulated by dexamethasone, as was GR itself. PGC-1α, required for glucocorticoid-induced maturation of primary mouse fetal cardiomyocytes in vitro, was also downregulated in fetal hearts at E17.5, 24 h after dexamethasone administration. Similarly, following a course of antenatal corticosteroids in a translational sheep model of preterm birth, both GR and PGC-1α were downregulated in heart. These data suggest that endogenous glucocorticoids support the perinatal switch to fatty acid oxidation in cardiomyocytes through changes in gene expression rather than gross changes in mitochondrial volume or mitochondrial turnover. Moreover, our data suggest that treatment with exogenous glucocorticoids may interfere with normal fetal heart maturation, possibly by downregulating GR. This has implications for clinical use of antenatal corticosteroids when preterm birth is considered a possibility. Key points: Glucocorticoids are steroid hormones that play a vital role in late pregnancy in maturing fetal organs, including the heart. In fetal cardiomyocytes in culture, glucocorticoids promote mitochondrial fatty acid oxidation, suggesting they facilitate the perinatal switch from carbohydrates to fatty acids as the predominant energy substrate. Administration of a synthetic glucocorticoid in late pregnancy in mice downregulates the glucocorticoid receptor and interferes with the normal increase in genes involved in fatty acid metabolism in the heart. In a sheep model of preterm birth, antenatal corticosteroids (synthetic glucocorticoid) downregulates the glucocorticoid receptor and the gene encoding PGC-1α, a master regulator of energy metabolism. These experiments suggest that administration of antenatal corticosteroids in anticipation of preterm delivery may interfere with fetal heart maturation by downregulating the ability to respond to glucocorticoids

    ELF5 suppresses estrogen sensitivity and underpins the acquisition of antiestrogen resistance in luminal breast cancer.

    We have previously shown that during pregnancy the E-twenty-six (ETS) transcription factor ELF5 directs the differentiation of mammary progenitor cells toward the estrogen receptor (ER)-negative and milk producing cell lineage, raising the possibility that ELF5 may suppress the estrogen sensitivity of breast cancers. To test this we constructed inducible models of ELF5 expression in ER positive luminal breast cancer cells and interrogated them using transcript profiling and chromatin immunoprecipitation of DNA followed by DNA sequencing (ChIP-Seq). ELF5 suppressed ER and FOXA1 expression and broadly suppressed ER-driven patterns of gene expression including sets of genes distinguishing the luminal molecular subtype. Direct transcriptional targets of ELF5, which included FOXA1, EGFR, and MYC, accurately classified a large cohort of breast cancers into their intrinsic molecular subtypes, predicted ER status with high precision, and defined groups with differential prognosis. Knockdown of ELF5 in basal breast cancer cell lines suppressed basal patterns of gene expression and produced a shift in molecular subtype toward the claudin-low and normal-like groups. Luminal breast cancer cells that acquired resistance to the antiestrogen Tamoxifen showed greatly elevated levels of ELF5 and its transcriptional signature, and became dependent on ELF5 for proliferation, compared to the parental cells. Thus ELF5 provides a key transcriptional determinant of breast cancer molecular subtype by suppression of estrogen sensitivity in luminal breast cancer cells and promotion of basal characteristics in basal breast cancer cells, an action that may be utilised to acquire antiestrogen resistance

    New genetic loci link adipose and insulin biology to body fat distribution.

    Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, here we conduct genome-wide association meta-analyses of traits related to waist and hip circumferences in up to 224,459 individuals. We identify 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (BMI), and an additional 19 loci newly associated with related waist and hip circumference measures (P < 5 × 10(-8)). In total, 20 of the 49 waist-to-hip ratio adjusted for BMI loci show significant sexual dimorphism, 19 of which display a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological mechanisms
